Eli Lilly Plans 2027 FDA Filing for Retatrutide After Phase 3 Weight-Loss Results

Eli Lilly said Thursday that it plans to seek regulatory approval for retatrutide, its next-generation obesity treatment, in the first quarter of 2027. The new filing timeline is later than the company's previous 2025 target.

LLY stock was up 1.99% on the day. The company said the delay reflects the additional time needed to collect and verify manufacturing and quality-control data required by regulators.

Retatrutide is a once-weekly injectable drug. Eli Lilly said it has passed two additional Phase 3 clinical trials, with weight-loss results that exceeded those of current treatments already available on the market.

In one trial, adults with obesity and Type 2 diabetes lost an average of 20.8% of their body weight over 80 weeks. That amounted to nearly 50 pounds on average.

In a second trial, adults with severe obesity and established cardiovascular disease lost an average of 22.6% of their body weight over the same 80-week period, or about 55.8 pounds.

How Retatrutide Differs From Existing Obesity Drugs

Retatrutide is designed to act on three hormones: GLP-1, GIP, and glucagon. That differentiates it from other major obesity medicines in the same market.

Lilly's current blockbuster obesity drug, Zepbound, targets GLP-1 and GIP. Novo Nordisk's Wegovy targets GLP-1 only. Retatrutide's three-hormone mechanism represents a newer approach in the obesity drug category. The addition of glucagon receptor agonism is associated with increased energy expenditure, which may contribute to the weight-loss magnitude seen in the trial data, though further regulatory review is needed.

Side effects reported in the retatrutide trials included diarrhea, nausea, and constipation. Those effects are consistent with other treatments in the GLP-1 drug class.

Kenneth Custer, president of Lilly Cardiometabolic Health, said the company believes it has the necessary data to file globally. He said Lilly is targeting not only obesity, but also knee osteoarthritis pain and obstructive sleep apnea.

Analyst Estimates and Wall Street Coverage

Analysts at TD Cowen estimated in January that retatrutide could reach $3.8 billion in sales by 2030. If achieved, that level of sales would make the drug a significant part of Lilly's obesity portfolio alongside Zepbound and its newly launched pill, Foundayo.

Lilly's development of retatrutide also comes as the company seeks to maintain its position against Novo Nordisk in the global weight-loss drug market, a category that has grown rapidly as GLP-1 therapies have expanded the patient population seeking pharmacological obesity treatment. The two-year filing delay narrows the window in which current treatments like Zepbound and Wegovy face limited direct competition from next-generation mechanisms.

Among 22 Wall Street analysts covering LLY, the consensus rating is Strong Buy. That view is based on 20 Buy recommendations and two Hold recommendations issued over the past three months.

The average price target for LLY is $1,294.06, implying roughly 17% upside from current levels.
